The reverse is completely blank, so it is almost impossible to tell for certain.

Coins like this were counter marked for a variety of reasons - often to extend their are of circulation either regionally (i.e. to prove that they can be used in the area) or temporally (i.e. to prove that they are still valid as current coinage).

Thank you for your ideas and comments. So can we say that the countermark was struck before the Romans conquered the Greek colony of Tomis or was this countermark also used during the Roman Empire?
